Dharmacon and Panomics, suppliers of QuantiGene® Reagent System, have jointly launched a genome-wide line of assays specifically designed to measure gene expression knockdowns by small interfering RNA (siRNA). Matching Dharmacon's human siGENOME™ to Panomics's QuantiGene Probe Set produces gene silencing assays for over 22,000 human genes.
The QuantiGene profiling products provide accurate and precise measurement of
siRNA-mediated knockdowns. When used together, Dharmacon's SMARTpool® siRNA products along with Panomics's QuantiGene Reagent System and Probe Sets enable researchers to accurately and efficiently validate RNAi gene knockdown studies supporting comparison and standardization among experiments and laboratories. The companies are jointly marketing and selling the assays to the life sciences market.

The QuantiGene Reagent System is based on branched DNA (bDNA) technology, an
FDA-approved, proprietary signal amplification method that offers robust and
precise RNA quantification directly from cell lysate or tissue homogenate.
When combined with the QuantiGene Reagent System the target-specific
Probe Sets (purchased separately) provide precisely controlled
specificity and signal amplification.

| Principle of QuantiGene
Assay |

Step 1
Cells are lysed to release mRNA in the presence of target probes. Target mRNA
from lysed cells is captured by hybridization and transferred to the Capture
Plate.

Step 2
Signal amplification is accomplished by hybridization of the bDNA Amplifier and
Label Probe.

Step 3
Addition of chemiluminescence substrate yields a QuantiGene signal that is
proportional to the amount of mRNA present in the sample.
