RNAi Discovery and Therapeutic Services

microRNA Profiling and Functional Characterization

microRNAs are emerging as not only key regulators of gene expression in fundamental cellular physiology but also as potential biomarkers in the characterization of normal biological processes as well as the diagnosis and prognosis of disease states. microRNA expression profiling, combined with phenotypic screens using libraries of microRNA mimetics and inhibitors, permits the identification and validation of microRNA function in both the pathology of disease and the profiling of responses to therapeutic intervention.

Our complete microRNA functional genomics platform combines robust and sensitive microRNA microarray expression profiling with phenotypic screening using Thermo Scientific miRIDIAN miRNA Mimic and Inhibitor libraries.

RDTS microRNA Functional Genomics Platform

microRNA Expression Profiling: From Samples to High-Quality Data

The RDTS lab provides global microRNA expression analysis from total RNA. The Thermo Scientific Dharmacon microRNA expression profiling platform utilizes a proprietary, two-color high-density 8-plex slide comprised of probes to capture all human, mouse and rat mature microRNAs in the miRBase database.

A Universal Reference for Each Experiment:

Thermo Scientific Dharmacon Synthetic microRNA Reference Probe Set is an equimolar mixture of 5’-Thermo Scientific DyLight 547-labeled, single-stranded synthetic miRNAs targeting all mature human, mouse, and rat miRNA sequences in the miRBase Database. The use of this internal reference provides reproducible data normalization and controls for variations due to up-stream processes (array manufacture, washing and hybridization processes, pipetting variation, etc). We use this proprietary microRNA Reference Probe Set as a standard reference in all miRNA expression profiling experiments.

  • Septuplicate probes in high-density format results in greater statistical power
  • Spike-in controls normalize for variations in labeling efficiency from sample to sample
  • Background controls correct for baseline fluorescence
  • Sensitive—sub-attomole detection limits
  • Robust data analysis—relative intensity data and error modeling using proprietary analysis software
  • Responsive service and proven expertise from the leader in small RNA biology
